Protective Effects of p-Coumaric Acid Isolated from Vaccinium bracteatum Thunb. Leaf Extract on Corticosterone-Induced Neurotoxicity in SH-SY5Y Cells and Primary Rat Cortical Neurons

Corticosterone (CORT)-induced oxidative stress and neurotoxicity can cause neuronal dysfunction mental disorders. In the present study, we investigated effects mechanism of HP-20 resin fraction water extract Vaccinium bracteatum leaves (NET-D1602) its bioactive compound p-coumaric acid on cell damage in SH-SY5Y cells primary culture rat cortical cells. NET-D1602 significantly improved viability CORT-induced cultures cells, increased activities antioxidant enzymes (superoxide dismutase catalase) against phosphorylation levels ERK1/2 cAMP response element-binding protein (CREB) neurons. addition, CREB by was dramatically reversed PKA, c-Raf/ERK, PI3K, mTOR inhibitors. Lastly, demonstrated neuroprotective (3 10 ?g/mL) ?M) via mediated ERK1/2, Akt, pathways. These results suggest that is a potential component NET-D1602, with ability to protect regulating mTOR-mediated phosphorylation.
